The Times’s Stylebook weighs in on the matter definitively, saying:

" Few besides New Yorkers stand or wait on line. In most of the English-speaking world, people stand in line. Use that wording."

I guess you're in the minority opus!

Standing in line you're part of the line. Standing on line, sounds like you're standing on the people in line.

I've always said stand IN line. But I have heard stand ON line too! I've heard everything just about, living where I live and English not being a first language with a lot of people. As a matter of fact my husband who's first language is Spanish says we have to make the line. And I had a Filipino friend that would say we have to fall in line! I always thought that was funny , I pictured people in the line falling down! hahahaha!

Another thing I think is funny, is laying in bed!! We actually aren't IN the bed, we are ON the bed. If we were In the bed we would be stuffed /stuck in the mattress or something right? Seriously, think about it! How come we don't say I'm ON the bed, not I'm IN the bed! ?

bring some paint with you, then you can paint your own line an proceed to stand ON it while you are IN the line.

or if you're British you can queue up for the queue.

Exactly. There can be a line on the floor you stand on. There can't be a line off the ground you're in.

The line that you are standing in is formed by people not by paint. So at the DMV or grocery store,etc. you are in a line of people. You are not standing on them/it.

Nah. Just today was a topic about a porn version of The Golden Girls, which I think got deleted, and somewhere you can find a topic about a fight with a waitress because toast and an english muffin were on same plate. So this is not even close
