All I can think of include the six kids on The Brady Bunch (specifically musical careers) and Al Lambert (Christine Lakin) on Step By Step.
Also worth mentioning are Laverne DeFazio and Shirley Feeney on Laverne and Shirley after they moved to California--they wanted to try showbiz besides working as gift wrappers at Bardwell's Department Store. Oh, and Carmine Ragusa (The Big Ragoo) definitely wanted to be in show business, both as a Las Vegas singer and on Broadway!
Didn't Joanie Cunningham on Happy Days have such aspirations?

Patty Lane in The Patty Duke Show wanted to in one episode, Mary in the Donna Reed Show once wanted to, and of course Lucy Ricardo and the Mertzes were always wanting to, Fred having already been in vaudeville years ago.

Jack McFarland on Will & Grace.
Not show business, but Ron Harris of Barney Miller and Diane Chambers of Cheers were aspiring writers who were eventually successful.
