The term originated in online gaming rooms as an acronym for We Owned the Other Team. It's basically just a victory call. After that it evolved into a general expression of joy, like "yay".

Although depending on whom you ask you might get a different answer.

it stands for "we owned the other team", i think initially referred by gamers; the o's are changed to 0's just for the hell of it, really. it's used in the same manner as "YAY!!" for anything though, not just winning a game.

haha I never knew it stood for anything, I always just thought it was "woo" with a 't' on the end because people wanted to sound cool

I had someone say woot to me once in a chat room, and I said bless you. LOL!! I'm not sure what it means either. Safety Pins explaination makes sense though.
