Bob Saget is best known for his eight years on Full House. This fall, Saget returns to playing a single father in the WB's new sitcom Raising Dad. But the comedian has even higher hopes for raising awareness about scleroderma.
His sister Gay passed away from the disease seven years ago.
"It was difficult for our family because she passed away so quickly," says Saget, former host of America's Funniest Home Videos. "It took just two years from diagnosis to when she passed away at age 47. But for her and others who suffer as she did, that time can seem interminable — you want to live, but the pain can be unbearable."
Scleroderma is a chronic, degenerative disorder that leads to blood vessel deterioration, tissue loss, and the overproduction of collagen in the body's connective tissues. Scleroderma is Greek for "hard skin," and patients often show a taut and leathery skin appearance due to the overproduction of collagen.
According to most sources, an estimated 300,000 Americans have scleroderma. Eighty percent of them are women of childbearing age.
Roughly half of the people afflicted with this disease will develop a limited scleroderma that is confined only to the skin.
The remaining scleroderma patients will develop systemic scleroderma, a life-threatening version that affects internal organs. Systemic scleroderma causes severe damage and serious complications for the body's digestive, respiratory, circulatory, and immune systems.
The cause of this autoimmune disease remains unknown. Scientists suspect that a genetic predisposition to the disease coupled with environmental factors trigger its development.
While pain control can make systemic scleroderma manageable, the disease remains incurable.
"And when we use the word 'manageable,' we mean only that the pain levels have been brought down to a level which allows the patient to get out of bed," says Saget, who volunteered to help the Scleroderma Research Foundation (SRF) two years before his sister showed symptoms of the disease.
"I was just astounded by the effects of the disease," says Saget. "It's relentless in its progression and in the elevation of pain. Because its victims are primarily young women, it tears families apart and leaves children alone, bewildered, and scared. And it has no cure."
Recognizing scleroderma
Symptoms of scleroderma may include:
Tightening, swelling, or thickening of the skin
Arthritis
Weight loss
Muscle, bone, or joint pain
Stiffness in the hands or feet
Skin discoloration
Shortness of breath
Hair loss
Swallowing difficulties
Heartburn and digestive problems
Calcinosis — White lumps under the skin that can erupt into chalky white fluid
Raynaud's phenomenon — Pain in hands; feet in response to hot and cold
Because many of these symptoms may be absent or may reflect entirely different conditions, diagnosis can be a challenge.
"It's not at all unusual for someone with some of the symptoms to see a few doctors before an accurate diagnosis is made," says Dr. Kari Connolly, associate professor of dermatology and medicine, University of California at San Francisco. "They can literally be wandering around for years."
That can delay much-needed treatment.
"Its true that there isn't yet a cure," says Connolly, "but we can and do treat the affected organs, and that does prolong lives."
There is no one standard test for scleroderma. And that sometimes makes diagnosis a challenge. Rheumatologists and specialized dermatologists like Connolly usually are left to uncover the disease.
Adding to the dilemma: The ANA, or anti-nuclear antibody test — a standard in making a diagnosis — may appear as a false negative in the early stages of the disease.
But a non-invasive test developed through SRF research funds can evaluate an early sign of scleroderma — poor capillary function. The nail fold capillary test examines the skin beneath the fingernail to determine vascular functioning.
Still, an array of diagnostic tests is needed to positively identify the disease. "What often happens is that the signs become more and more obvious," says Connolly, "and then the diagnosis is finally made."
"Then the other shoe falls," adds Connolly. "They find out there isn't a cure and they may just walk away."
"That's really a sad part of this process," says Sharon Monsky, SRF founder and chairman of its board of directors.
The mother of three kids, Monsky has lived with diffuse systemic scleroderma for nearly 16 years. "You can live a full and rich life, but you have to participate in the process." That process includes a wide variety of treatments to bolster failing organs, boost immune function, and decrease pain.
Adding to the confusion is "the wide variety of treatments given to patients in the advanced stages of the disease," says Saget. "I watched my sister suffer some side effects that were horrific. I later found that these were experimental treatments that did nothing to alleviate her suffering."
"But when you find out you have the disease you're likely confronting something you've probably never even heard of, and that means we have a steep learning curve to find out what the disease is all about and which doctors work well with it," says Saget. "Not all do."
Saget suggests contacting families with members who have the disease or using the physician locator from SRF.
Future hope
Hoping to accelerate finding a cure for scleroderma, the SRF is working closely with four university-based research centers to "share both our research successes and our failures," says Monsky.
Recent successes from this multi-institutional approach include:
The autocrine loop — "Our research strongly suggests that fibrosis is a self-sustaining process built on an autocrine loop," says Monsky. "It's a self-feeding positive feedback cycle." Interrupt that cycle and disease progression might be interrupted.
Microvascular dynamics — Capillary tissue damage has long been thought to be a byproduct of scleroderma. But new research indicates that these vascular dynamics may actually play a central role in the disease.
Antibody clustering — Antibodies that cluster at the same cellular location and are unique to scleroderma were reported in the Journal of Experimental Medicine. This "points the way to identifying the environmental factors which lead to disease development, as well as helping to eventually isolate genes involved in its predisposition," says Monsky.
For example, "we're discovering how the skin is changing on the molecular level," says Connolly, "and that was somewhat a mystery."
Connolly is also directing the assembly of an extensive tissue bank devoted exclusively to scleroderma. "As this grows it allows us to pull out tissue samples from various stages of growth and find out what is actually going on, rather than work on hypotheses developed from more limited observation. We're already finding some unusual cells that were quite unexpected."
Coupled with vast amounts of computer time and expertise donated by Silicon Valley biotech firms, these forays "allow us to cluster the data and develop meaningful expectations for the next 6-12 months. We're no longer limited to a few hypotheses."
"It's going to be a curable disease," says Saget. "But we've got to support the research because there's still a long way to go."

John Stamos" (born August 19, 1963) is an American actor and the son of Bill and Loretta Stamos.His paternal grandfather had borne the name ''Stamotopolous'', and changed it to ''Stamos'' on immigrating from Greece to America.
Born in Cypress, California, John has two sisters, Janeen and Alaina. He has appeared in a number of movies as well as television sitcoms since 1982.He kicked off his acting career with a role in ''General Hospital'', though he starred in a Coke ad which was never aired.
John played the character of "Jesse (Hermes) Katsopolis" in ''Full House'' (1987-1995), an "Elvis-obsessed" rock and motorbike freak who moves in with brother-in-law Danny Tanner to help him raise his kids (Donna Jo ("D.J."), Stephanie and Michelle) after the death of Danny's wife Pamela. Jesse has his own rock band called "Jesse and the Rippers." He is basically a "cool and carefree" guy who is a hot favorite amongst the opposite sex, he has great hair, which he pampers a lot all the time! Later he marries Rebecca Donaldson, who is Danny's beautiful co-host on "Wake-Up-San Francisco," and has twins: Nicky and Alex.

Dave Coulier was born on September 21, 1959, in Detroit, Michigan, and was raised in the nearby suburb of St. Claire Shores. Comedy wasn't Dave's first goal in life--the hockey net was. As a teenager, he was a star of his high school hockey team. He was talented enough to dream of a pro hockey career. An avid athlete, the broadly built young man also excelled in water skiing and swimming.
But at the same time, Coulier was discovering he had strange voices inside himself! And these voices and impresonations tended to pop out at odd moments, to the delight and amusement of his teammates and friends. While Dave was still a teen, his comic routines brought him his first comedy job--performing (and writing) commercial voice-overs at a local radio station.
With his great love of both sports and comedy, Coulier's career was at a crossroads. As it turned out, the turning point came at Christmas, when Dave was just 17. During a hockey game, he was bringing the puck out from behind his own net when suddenly, out of nowhere, an opposing player slammed into him. "I woke up in the hospital," remembers Coulier, "and they were stitching my face up. I remember seeing a needle and thread being pulled from my face. My bones were broken. There was quite a bit of blood. My dad said, "You're pretty banged up."
This story is a rare thing for Coulier--it's not funny. The accident left Dave with a permanent scar. It also helped him make up his mind. Right then and there, Dave decided that making silly noises, doing nutty voices, and making people laugh would be a better--and safer--way to make a living! Dave started doing standup in local Detroit clubs. At 19, he took the plunge and moved to Los Angeles. There he became an overnight success and lived happily ever after ... Not!
Dave remembers an eight-month stretch when he was too broke to afford transportation around Los Angeles. It had been eight months since his last standup job. That Thanksgiving was a lonely one. He couldn't afford to fly home to Michigan to be with his family. Dave's life was following his character's, Joey Gladstone, but without the laughs.
On the other hand, Dave had the support of his best friend to help him through the rough times. Just as in the show, his best friend was Bob Saget. "We've always been like brothers--I say he's the sister I never had," says Saget. "I've even had a dinner at his dad's house in Detroit and slept in his old room. Once, Dave stayed on my couch for three weeks. He'd pretend he was my bizarre nephew, Busby, and do funny voices."
Finally, Dave got his first big break. I wasn't exactly glamorous. He landed the job of doorman at the famed Los Angeles comedy club, the Comedy Store. He soon worked his way onto the stage and began collecting the gags and funny characters that are his trademarks. This led to appearances on Showtime's Laff-a-Thon, and HBO's Detroit Comedy Jam. Thanks to the funny voices that had made him a class clown, the young comedian started getting work on Saturday morning cartoons. He twisted his vocal chords around such character voices as Scooby Doo and Mork in the cartoon version of Mork and Mindy. If you tune in to Muppet Babies some time, see if you can recognize Dave's voice as Animal and Bunsen Honeydew. His colorful vocal performances won him an Emmy!
By this time, Dave was getting to be well known in the business. Now he not only had money to travel, he was on the road 42 weeks out of the year, performing at comedy clubs across the nation. Then, in 1987, he landed his starring role on Full House and left standup behind, without regrets. "Standup is a grind, man," comments Dave. "It's the hardest thing I've done! You never know when you're going to die one night in front of 300 people. On a sitcom, you can always say, "The writers didn't come up with the right joke there!"
Does he mind competing for laughs with his old friend and fellow standup comic co-star, Bob Saget? "We're best friends," Coulier says firmly. "There's no jealousy on our show." Saget agrees. "He makes me laugh more than anyone else. He's the kind of guy who sticks his head out the car window and yells something and it's funny and you don't mind."

Scott Weinger
Scott Eric Weinger (pronounced, btw, as wine-gerr) was born in New York, New York, on October 5, 1975, to Babs Weinger, a teacher, and Elliott Weinger, an orthopedic surgeon. The eldest of four children, Scott has two brothers and one sister. He spent the majority of his formative years in southern Florida, then moved with his family to Los Angeles when his career began to take off.
Scott first became interested in acting in the third grade, when an actor gave a presentation for Career Day. He relentlessly pestered his parents to get him an agent until they finally realised that the young tyke was serious about becoming an actor. His first gig was a national commercial for Ideal Toys.
In autumn of 1994, Scott left the L.A. scene to fulfill yet another dream - attending Harvard University. Taking leave of TV series "FULL HOUSE" didn't suggest he was leaving the business altogether. He continued as the voice of "Aladdin" in the Saturday morning TV series, completed two more full length Aladdin videos, and made a final appearance on "FULL HOUSE", all while maintaining excellent scores at school. As if all of that didn't keep the ambitious lad busy enough, he also held a part-time job as a youth correspondent for Good Morning America.
Scott majored in English and minored in French literature while at Harvard, and he graduated magna cum laude in June of 1998. In his first online interview after returning to Los Angeles, he still seemed to be undecided about what his plans were. The possibilities were narrowed down to writing, directing, acting, and news correspondence.
Since his return to Hollywood, he has starred in a horror flick, SHREDDER, produced an award winning film short, "The Cricket Player", and provided his voice for Osamu Tezuka's METROPOLIS, Disney/Square Co.'s video game, Kingdom Hearts, and two upcoming Disney films, MICKEY'S PHILHARMAGIC & THE SEARCH FOR MICKEY MOUSE!
Scott considers himself to be a writer, primarily, and an actor, secondarily. He is currently working as a writer for the WB television show, "Like Family," which is described as a "multi-ethnic crossover comedy about two very different families coming together under one roof."

Ewww.... John does NOT look good with that beard. I think he looked a lot better during seasons two through eight on Full House! Maybe it's just me, but I don't really like so much of a bearded look on men. :rolleyes:
